t(15;17). Composite illustration of the morphology and cytochemistry of APML. |
1. MGG. Hypergranular M3. Bilobed nuclei partially obscured by abundant purple granules. 2. MGG. Promyelocyte packed with Auer rods, partially obscuring the nucleus. 3. MGG. Hypogranular promyelocytes with bilobed nuclei (centre), with some moderately granular promyelocytes (M3 variant). 4 & 5. SBB. Typical heavy Sudanophilia filling the cytoplasm and overlapping the nucleus. 6. MPO. Typical heavy staining of 3 promyelocytes. Auer rods visible in the left-hand cell. 7. PAS. Typical diffuse cytoplasmic blush due to acid glycoproteins in the granules. 8. Combined esterase. Typical chloroacetate positivity (blue) in all the promyelocytes, with positive Auer rods. 9. Combined esterase. Aberrant expression of monocytic esterase (alpha-naphthyl butyrate, dark green/brown) in two promyelocytes, with usual blue chloroacetate positivity in two cells. Note chloroacetate positive Auer rods in bottom left butyrate positive cell. |
